Darren Wilson, 35, of Ballarat appeared before court on Saturday night charged with one count of murder.
He was remanded in custody to appear at Ballarat Magistrates Court on Monday.
Police earlier said the teenager and a man were taken to a house on Carlyle Street, Scarsdale, by a 16-year-old girl and a 19-year-old woman who lived there.
They were confronted by a group of people, and a fight broke out about 5am (AEDT) on Saturday.
During the altercation, the boy was struck with a blunt object and suffered a serious head injury.
Paramedics tried to revive the youth, but he died at the scene.
Police arrested two men and a woman on Saturday afternoon.
A 39-year-old Wendouree man is in custody assisting police with their inquiries.
A 28-year-old Ballarat woman has been released pending further inquiries.
